I like Alaska enough that I infrequently fly up for a 3 day weekend.....but with that obvious bias aside, I'd second some of the previous thoughts to suggest an AMHS trip through Southeast.
Recognizing that it can be expensive (particularly with the cost of xporting a vehicle), the ratio of effort to Spectacular in Southeast is pretty low. Just going for a ride on the ferry takes one through some of the most scenic sights in Alaska, not to mention what can be gained by a flightseeing trip from one of the SE towns.
For full disclosure, I've never taken the inside passage, but have been to most places in SE (Juneau, K-town, Sitka, Wrangell, Kake, Craig, Petersburg, Yakutat, etc) and while I don't believe that SE is the only place to see in Alaska, it is one of the more scenic areas and with a unique ambience. Just bring good rain gear.
