I-81 is a good (if obvious) option. I think I have mostly gone via I-75 to Knoxville, though sometimes I-77 and I-26. Both work and miss most of the worst of the cities. (Atlanta on I-75 is an exception, though I've usually not had an unreasonable time going around it.)
If you have plenty of time, the Blue Ridge Parkway to Skyline Drive is wonderful. Don't be too put off by the low tunnels; they are all barrel-shaped, and the heights are measured at the shoulder—the middle is tall enough to easily clear a typical RV if you keep fairly tight to the middle line.
