Ft. Pierce is the easiest as from there I95 goes out to the west a bit, and the Turnpike is a more direct route. That intersection also has a decent selection of restaurants and service stations.
You can go down I-95 to Jupiter if you want to save a little bit of tolls - that's the next place the Turnpike comes close by. Both exits are well marked.
We like to stop for diesel at the last service plaza before the Keys (about mile marker 19) because it is easier in-and-out than the stations once you get off the Turnpike ....
