If the kids like outdoor stuff So.IL would be good, lots af waterfalls, burdenfalls, jackson falls, trigg tower, bellsmith springs, dixon springs, garden of the Gods, pounds hollow, giant city park, (good food at the lodge ).
If they are not into outdoors, St.Louis would be the ticket, lots of things to do go up in the arch if you go there, and lots and lots of good food, Cohokia mounds is an awesome spot for kids, six flags, graints farm, the science center is a must for smaller kids.
You could also spend 1/2 the time in one place and move to the other as they are only 2 1/2 hours apart.
