Given similar design there is no difference between a single 12V and two 6V. Internal construction between 6 and 12V on Lifeline batteries is the same I can not speak for the major battery manufacturers. Important to remember is that all batteries need to be the same age and condition or the system will be dragged down to the weakest link.
