What is the best route to take from Dayton, Ohio I-75? Also some nice places to stay along the way. We plan on leaving on Saturday possibly Sunday and arriving on Monday for check-in time at 3:00pm. T...

I-75 would be my choice. Take I-75 South to Florida’s Turnpike South (exit 328, old 65), to I-4 West (exit 259) to Walt Disney World Exits. I would leave Saturday.
We like Holiday Travel Park in Chattanooga TN (Rossville GA) about 6 hours from Dayton with no stops.
We stop in Lake Park GA at Eagles Roost or there is a remodeled campground Good Sam at Camping World across the highway. Lower rates. Good for overnight. There is a Flying J at Lake Park also. Lake Park is about 5 hours from Chattanooga with no stops.
Then get on Florida Turnpike at Wildwood (toll). $10.11 for 4 axles. I would stop and get a SunPass transponder and set up an account. Mini transponder $4.99. You can purchase one via vending machine at:
