Second question first: the interior lights run off the house battery. Basically, the only things that run off the starter/chassis battery are things directly related to the chassis: the main engine starter, the transmission, the headlights and running lights, that sort of thing. Often a power entry step is also wired to the chassis battery; and occasionally the generator starter is wired to it as well. Interior lights (possibly excluding a light or two in the cockpit area), appliances, etc. all run off the house battery.
First question: You have to follow the wires to see where they lead; unfortunately, there's no simple foolproof way to figure such things out without doing that work. Wires that lead directly to a chassis ground connection (perhaps via a shunt if you have a fancy battery monitor) are obviously negative ones. Wires that lead to a fuse or circuit breaker are positive ones. Hopefully there aren't too many others to sort out.
