No, DEF is not required in older DP motorhomes. I just filled our DEF tank for the first time and we drove about 2,300 miles on 13.771 gallons from a 16 gallon DEF tank.
I think it works out to one tank of DEF to 4 tanks of fuel. We have a 230 gallon fuel tank.

On Edit: If you put DEF in the diesel tank, it will ruin the engine within minutes and the whole fuel system will need to be replaced. It's pretty hard to confuse the diesel handle and the DEF handle but it has happened and is an expensive mistake.
