I've been doing long drives, more than 1000 miles a day when younger, for 50 years. The best thing for making this comfortable over the years has been making sure that the seating position fits my body (critical at buying time) and making stops to get out, move around, relax my muscles. Arms, legs, back, abdomen, neck muscles all get fatigued trying to hold a driving position.
In my 20's, I could do 5-6 hours non-stop, 4-5 hours between stops on long drives. This fit pretty well with vehicle range on what I was driving at the time. Into my 50's, this got down to 2-3 hours between stops to get out and move around. Now that is my max, I might go that far between stops at the beginning of the day, but I might be stopping every 30-60 minutes after 8-10 hours of driving, which is a good indication I need to stop for the night.
For what it is worth, the strictly enforced (by electronic monitoring) standard for commercial drivers in the European Union is a 30 minute stop after two hours driving. My stops are more like 5-10 minutes, 30 would be a meal. While it might mean passing again trucks I passed earlier in the day, these frequent short stops have little impact on my travel for the day, maybe slightly slower average speed but usually I can go farther before hitting a limit.
