FIRST....stop putting 80-100# into your rig even the flush connection. You will blow out the anti-siphon/back flow preventor and cause a big flood inside your rv.
And if you don't blow it out you will probably blow a fitting off on tank sprayer and make it useless.....50# MAX is all you need. Use a water pressure regulator
As for the 'stuff' growing in tank....hummmmmm
Could be fungus, could be mold, could be scum.....just need to fill tank full and add bleach (1/4 cup per 15 gallons) and let it sit overnight then dump/rinse tank.
Oh.I take it that vent line is now clear
