Many years ago I read there is almost no difference. One study - Motortrend maybe? - found that a small amount of "vacuum" formed behind trucks cabs when the tailgate is down. Actually found there was a miniscule increase in mileage, meaning better mileage, when the tailgate is up. The best they could figure is that the up tailgate directed some air into the "vacuum", breaking it up, and increasing fuel mileage. All numbers, though, were very small - realistically no difference.
I haven't read or seen anything recently.
