You have 50' of open pipe/hose from RV to the barn with the RV's valve open?

My guess would be that you have the dreaded pile-of-petrified-poo in the tank, your tank is full or blocked, and that nothing is reaching the drain pipe.
That was my first thought. Or a venting problem.
To prove once and for all that the sewer is OK, close the drain valve. THIS IS IMPORTANT: :E Disconnect the line at the trailer, (be prepared for a catastrophic leak; ready to reconnect really quickly) :E again!
Then take a water hose and run lots of water down your sewer line. If it drains well then it's fine and the problem is the trailer.
Next, place a bucket or something to catch what may be in the tank and s l o w l y open the drain valve, a bit at a time.
When everything has drained out, hopefully not too much--if the tank wants to drain a lot, close the valve and reconnect to the sewer and drain the tank.
Then with the drain disconnected again, open the drain valve and slowly run some water into the toilet and see what happens. Does it run right out?
Let us know the results. Then you will get more suggestions.
One thing comes to mind is venting. If there is no way for the air that is displaced by flushing the toilet to escape strange things can happen. I won't get into that yet, let's see what you find first.
