That connection is for washing out the tank itself. It is unlikely that it's effect will reach up into the plumbing. When you do use it to wash out the tank, ensure the drain valve is open before you start.

They make a short hose with small holes on the end that attaches to a garden hose that you can stick into the toilet opening that sprays to wash out obstructions. I would run it up and down to try to get the bowl to drain before using any addition water.

In a pinch, I once used the end of a spare water hose that I ran up and down to create an opening into the tank. Once that was done, I turned on the water to give a good rince from the bowl.

Not all 5th wheel toilets make a straight drop. If the toilet is over the "garage" you might see a slight bend in the pipe which may allow some waste to hang up and create a clog. Dilution is the solution and sometime a little mechanical agitation is required from time to time. After making a "large deposit" it does not hurt to fill the bowl with water for a good flush to ensure everything clears the bend.

If you have a "snake' you might send it down the line just be very careful not to damage the bowl valve and seat.

Normally RV toilets drop stuff strait down into the tank and have no corners in between. As a last resort one could shove a stick down there (have a garbage bag ready to put it in) and try to break "things" up.
Make sure after this that she know not to keep the valve open and to use LOTS of water.

Fill the bowl then shut off the water supply so the bowl does not over fill and open the dump valve. Give it a minute for the weight of water to move the blockage into the black tank. If no move find something to poke about in the drain below the toilet while holding the toilet dump valve open. The drain line from the toilet to the tank has no traps etc in it maybe a bend or two.

Water does enter the bowl.
