I've done 2 long trips now towing the camper, bother over 2k miles. One was from KS to DC through the Appalachians and the other from KS to TX.
On the trip through the mountains, I locked out 6th once I was in the mountains. On the trip to TX I stayed in 6th at 1400 rpms at 65mph 98% of the trip.
Our camper only weighs 8500lbs loaded to camp, but it really is a dream to tow! The trip through the mountains and back I managed about 11.5 mpg.
I always travel at 65mph unless the speed limit is lower. Very rarely will I bump the speed up in city traffic if the flow of traffic dictates it. Do please note that this MPG was definitely ideal, as I usually average 11-12 mpg. I was the benefactor of some nice tailwinds on the way home on this trip.
If your camper is around the same as mine, I wouldn't lock 6th out unless its a very hilly area. I just play it by ear and if I see it shifting on every hill, I lock 6th out.
