Is there any particular rule of thumb, so to speak, where a person should do this?
Speed limit while loosened?? Common sense of course, but is there one? (speed limit that is, I'm already past the common sense limit. :B )
Any other factors to be considered when running tie downs loosened for trail purposes???

There really isn't any hard set of rules for "how much to loosen tie downs" On my rig I have Hapijack system front and rear, these have served me very well over the years and on some pretty good off road adventures.
You'll have to experiment to find what does work with your rig, even a change in suspension, tire pressure,frame flex how much you have loaded up your camper and air bags will have a effect on adjusting your tie downs setting.
Example, before our last trip to Anza Borrego to meet up with anutami I had an extra contact overloads added and a 40 gallon fuel tank install.These changes had the results in smoothing out some of the frame flex of my Ford and I didn't have to loosen my tie downs as much as I use too.
Things to try and all these suggestion will have effect on loosen your tie downs from minor to very loose (almost falling off)
1) Tire pressure.
2) Adding/ subtracting suspension components.
3) Air bags pressure settings.(if you have them)
4) How loaded up the camper is.
5) How/what kind of road your on.
6) frame flex
7) How big your TC is compared to the truck hauling it.
