There's probably no clear cut answer.
Manufacturers testing for EPA certification done on dynamometers. CR, which I'm no fan, of actually drives the vehicles. Can be argued that differences in test driver style/behavior make the difference, but I once read CR's description of their test procedures/protocols. Looked like they do all they can to keep the testing uniform.
