We actually were on a big trip, Niagara was our final northern destination, then many other sights on the way back down. Northern NJ (where I grew up) used to take me about 18 hours from tampa, but that is going the I-95 route. It took us about 6 hours additional to get to Liberty State park from Niagara. So, you should be looking at 25-26 hours from the border.
Dont know about the other route.
