A "web-based" client such as Gmail is one where your web browser interprets code sent from the server and presents you with the screen and with the data, in this case your email messages. These web-based clients only work with an Internet connection.
A PC-based client such as Outlook, Thunderbird, etc. is an application program that runs on your PC generally without the aid of your browser (although it could use your browser). The PC-based client sends requests to the server that result in sending and receiving emails, but not the screens that display them. These clients allow you to store copies of your email in local folders on your PC that are accessible when you are not connected to the Internet.
The email protocol does not actually require the Internet to work, messages may be created and sent over many types of networks using the same transport protocols used over the Internet.
