There will be a huge difference while towing. You won't feel the rear of the truck cycle up and down, up and down as much when going over rolling type sections of roads or sections like around bridges where they connect to the regular road and there is a height difference.
I usually measure my rear bumper height with no trailer connected and then fill the airbags to get back to that height once the trailer is connected. Not sure if this is correct but truck and trailer look correct (level) and goes down the road just fine with no bad manors.
