It depends on what you're interested in. driving across 41, you have Big Cypress Visitor Center near Shark Valley. At Shark Valley, you can either take a 2 hour Tram or ride your Bicycles on a 15 miles Loop. They sometimes do a Full Moon Bike Ride at 5:30 pm. Amazing number of birds nesting above the alligators.
From the Coe Visitor Center to Flamingo Campground is about 40 minute ride. So stop at Visitor Center on your way in.
Long Pine with the Anhinga Trail/Boardwalk is where you see the majority of alligators but lots of birds at other places.
