Sounds like one leg of the trip may well take you near St. Louis. With kids assuming they are not toddlers, I'd suggest taking a day at the St Louis City Museum. This would be doubly good if it happens to be a leg where you're otherwise driving straight through, as it breaks up the long span a little bit.
The City Museum is an entirely unique place, more a giant playground/jungle gym/interactive art project as a museum in the traditional sense.
The Ohio Turnpike has a few rest areas with overnight RV facilities at a reasonable price. If you end up with an overnight in that general area, one of them might work out very well. I would not complain at all if more states did something like that.
