Looks like a very do-able trip. We have visted every one of the stops you mentioned, but not all on the same trip.
We made a 3000 mile trip in 2012 which included Niagara Falls and Charlottsville. But then I was younger, only 79. :)
Last Summer we made a western trip of over 5000 miles, so we are not slowing down much.
Congratulations to your parents. That trip in 2012 was our very first time to visit Niagara Falls so we called that our 55th anniversary honeymoon.
