No. How's that going to happen?  It's not like black water somehow climbs up the tube and swims upstream to the faucet.
Actually, it can happen.  If there were no one-way valve on the connection, the water flow back at the spigot to the fresh water tank could create a vacuum and siphon the water from he black tank (if it was full) back into the fresh water valve.  That is exactly why the mfg'ers put the one-way valve on there.
I also use a "Y" at the spigot and use two hoses.  White for fresh water tank, green for black water tank.  However, I put quick disconnects on the ends of my hoses and in addition to turning off the water at the "Y" to the black tank, I will also remove the hose from the "Y".  I once forgot to turn the water off and had a huge mess to clean up.  :E
