I think that the only option offered for the E-450 is 4.56:1, and it's been that way more or less forever. Maybe aftermarket or kludging could get something else, but I personally don't think it would be worth the effort.
The newer six speed transmission would be worth looking for; the gaps between the top three gears on the five speed (and the identical gaps on the older 4 speed's top three gears) are rather on the wide side.
