I don't know about your RAM, but with my F250 tow/haul does not lockout overdrive, it just waits longer until shifting into it.
For highway driving I don't think you will see much difference in fuel mileage with or without T/H mode.
Around town driving, you will find that T/H may make the truck "feel" more powerful with quicker shifting, and the engine braking is nice when you get used to it.
When in doubt, use Tow/Haul.
