Can't answer specifically as it relates to an RV's roof but can from a building's: mold can start immediately depending on the local atmosphere. Keep in mind that just because the immediate surface has dried doesn't mean that water trapped further down has. And water always travels down seeking the path of least resistance. I'd work hard to do a total roof repair as soon as possible.
