I use Mapquest, DeLorme Street Atlas (within Topo) and Streets and Trips.
For the early "what if" planning considering alternative routes, different stops (overnight and sightseeing) I still prefer Streets and Trips for its flexibility and ease of use, ease of changing plans and saving alternatives.
The POI database, particularly RV parks and campgrounds, is not as complete as I would like, but I use a campground directory (Woodalls and/or Trailer Life, now replaced by Good Sams) to fill in, and a stack of road trip books and state or regional travel guides to find what is really interesting to me.
If you are not doing "what if" and want only one answer, Mapquest works pretty well.
