It's plausible that the valve stem could be plugged, I'd imagine particularly if there was something along the lines of fix-a-flat put in at some point.
If it were my problem, I'd probably remove the wheel (so as to get access) and see if I couldn't get things working by removing and replacing the valve core. If the air won't come out when the valve core is removed, it's probably time to go to a tire shop, I'd guess. It may be possible to free up whatever is causing problems by poking at it with a wire or something, but that strikes me as potentially a pretty dangerous stunt to try.
