I've done it several times. First time (for a long trip, anyway) DW and I went across US 2 from the east coast to the Olympic Peninsula, then she flew home and I went south along the coast to LA, then back east.
The second time, I towed from the East Coast to Las Vegas for a professional conference, she flew to meet me, and we circled the Grand Canyon and hit Bryce and Zion before she flew home, and I dragged the TT back east alone.
She has NEVER helped me hitch up. I've gotten pretty good at it by myself, and only recently got a new TV that has a backup camera. It is slightly helpful. DW has also never been particularly useful backing into a site. She's prone to stand where I can't see her, and when she doesn't do that, she gesticulates wildly -- I've never been able to convince her either that she's not much help gesturing when I can't see her, or that nonsensical hand waves are not informative. So to save the fight, I let her stand where ever she wants and wave her arms all she wants, and I back it in while ignoring her.
So, yeah, i tow the TT alone without fear. You can do it too.
