Many RV shops will smear additional sealant over the old stuff (which was smeared on the outside and cracked), and some recommend doing this every year. My opinion is that while this might be a very temporary fix, it isn't the way to go unless you have the camper for sale :). A roof vent or door, properly sealed with polyurethane, should be good for at least 10 years if not 20, with no attention other than occasional inspection. The only exceptions I have seen is the vent jacks, which seem to be particularly floppy. Even on those 5-10 years is a reasonable expectation.
Some time ago I posted a thread
here about how I would do this. Sealant spread around outside the flange does very little good. You will not see that done on any boat for example. It is the sealant between the flange and the surface that is doing the work, and that is not accessible without removing the door/vent/etc.
