The cover is a good idea. I've used one for 10 years and credit it for my TT still looking like new -including the graphics. It keeps my Dri-Z-Air from filling up as often too. Without the cover I had to dump them three-plus times during the winter, with the cover it's just over once.
No to the vent being open. The tighter you seal up the RV the less dampness you'll have.
