Having drove through there, towing I would stay on the interstates. Either catch 77 up to 81 and down to Bristol/Blountville exit, or take 40 to 26 in Asheville and take 11E in Johnson City to Bluff City. If you are staying at Lake View in Bluff City, nice park.
