Yes, you would need to locate and drill the proper sized center hole. Unfortunately, it looks like you are space-constrained by the size and placement of the cabinet stiles and back rail. I don't think you'll have enough space to locate that center hole without modifying the cabinet somehow.
The tool used to tighten and loosen the under-cabinet water fittings is called a "basin wrench", available at big box and hardware stores.

Well, I got real lucky. Made my mark and went for it. Used a 1 1/4" hole saw. Which was exactly the same size as the center portion of the faucet. Was a little too tight. Got out the dremel with the barrel sander and made the hole just a little bigger.
When I slid the faucet into place, the stile on the cabinet was VERY close. So close I had to have the DW hold the faucet up so I could get the 2 washers and lock washer on above the stile of the cabinet. It worked! I also added a 1/4 turn valve on each line. Thanks to who ever suggested that.
Now I have a cramp in my knee from the strange position I was in for way too long!
