My V10 E350 has been great! I do have 4.56 gears out of the E450 plus the Banks PowerPack. My combined weight is typically 22,000 pounds. Most of my towing is in the Cascades. I have no problem exceeding the speed limit going up mountain passes with a full load.
I'm coming up on 415,000 miles, and no major emergency repairs on engine or transmission. I did decide to do injectors and the timing chain preventively, because mine gets used way harder than most. Over half those miles are towing miles.
