Most folks here will generally say it is OK that "not secured" to the truck is fine (assuming it is in the truck itself.) A simple test as member Tracking describes is one way to test for static point loads. I would be careful thought to makes sure no dents to the roof of the cab could happen if something goes awry.
If you ask "not secured to the truck" meaning off the truck, then again, generally folks might say yes. You need to make sure that the mfgr's does not require full support for say the domestic water tank- you don not want that falling out of the floor. Camper makers may even state if their TC is safe for off camper use.
Your profile photo make me want to ask more specifics of "not secured." I would not do it if it stored as shown in that photo. Quite honestly, I would be a little wary of even walking inside let alone getting in the upper cab. The natural gravitational forces seem to be supported OK, but I see too many hinges in your set-up. Stacked block with only 2 of the old style jacks would not make me comfortable (and I am about one 6-pack away of being your stated weight.) If you are asking "unsecured" for that photo in your profile I would say no. For my comfort level I want to be sure things are stable- JMHO. If you are not referring to your profile photo then my apologies for assuming.
I use 5 tiers of PT 6x6: There is 3/4PT attached to the bottom of the camper and 2 6x's - continuous in the long direction tiers, 3 each in the short direction. This is to fully support my camper when off the truck at home (I never take it off in the field.)
The lumber takes almost all of the weight as the jacks are there just to keep it all steady... just a crank or 2 on the jack the hit the ground. I feel this is very stable event though it it relatively tall. I do not need to go much higher when loading it on to the truck.
