I like 3M products and I find that the adhesive backing they have on their Velcro is pretty strong compared to no-name cheaper ones. Just prepare both surfaces properly (I clean it with rubbing alcohol) and it stays on well even after constant use.
I did something similar to cover a bedroom roof vent, except I didn't run the Velcro all the way around because I wasn't worried about gaps:


I think with an even bigger strip of Velcro, there's even less chance of it coming off the wood trim. Also, my personal preference is to put the "hook" side on the removable part and the "loop" side on the permanent side.
(BTW, I have no relationship with 3M so there's no ulterior motive for me to promote them other than the fact that it works for me.)
Instead of using Velcro, some kind of clamping or hook system that would hold the Plexiglas flat against the trim should work, too. A roll of magnet strip like this is possible, too:
