There's no reason to replace the powertrain on a preventative basis. I replaced my V10 at 458,000 miles, because a few thousand before that I burned a valve seat by pushing onward with a plugged fuel filter an an aggressive tune (that I couldn't restore because the tuner took a bath that same weekend). When I scoped the engine, the hash-marks were still visible, and besides the one damaged valve seat, compression was good. Previous relative compression tests were all within factory spec. I was prepared to just have the affected head re-conditioned, but book time included pulling the motor, so at that point I decided to get a factory reman.
FWIW - My 4R100 is original, now with 489,000 miles on it. Majority of those were towing miles.
What I would consider replacing on a preventative basis is the upper radiator hose, and the heater hose that comes out of the intake. These both take most of the heat, so they get weak first. Also check the hard lines going to the rear HVAC. These can corrode in salt states.
