It depends on I95 traffic. If it is moving along, stay on it as long as possible. If it is slow going, get off and take the backroads, at least for a few miles around the traffic jam. Just an FYI, from exit 33 to Santee you can jump on Hwy 15. From Santee north to Exit 134, you can jump on Hwy 301.
Hwy 501 is heavier traffic but dual lane road. Hwy 378 is a nice road through some pretty sections but single lane road.
Another option if you have the time is to jump off on Hwy 17 taking it up to through Charleston and Georgetown. Will be a bit longer, but much nicer now that the have the section from Beaufort to Jacksonboro dualed. If you want to go this route, split the trip up into two days by staying at the Oaks CG south of Charleston.
