Hwy 501 is heavier traffic but dual lane road. Hwy 378 is a nice road through some pretty sections but single lane road.
Another option if you have the time is to jump off on Hwy 17 taking it up to through Charleston and Georgetown. Will be a bit longer, but much nicer now that the have the section from Beaufort to Jacksonboro dualed. If you want to go this route, split the trip up into two days by staying at the Oaks CG south of Charleston.
This is our usual route... But I hate the 17 traffic closer to Murrells Inlet and north.... That's why I thought of staying on 95 longer...
