Agree on 41 to Krome and then south to US 1. As to the poster that suggests that I75 would be faster for your route, that would be wrong. From Club Naples, I 75 would dump you off on between N. Miami and Ft Lauderdale. You would have, at anytime of day, a good drive from there just to get as far south as to where US41 would take you at Krome.

I'd definitely go 41, but I am on the east trail.
It's a straight shot to Miami, then to Homestead and the keys.

I-75 is faster but 41 is more scenic. You can also stop at the Shark Valley Everglades NP entrance to take the tram ride, it is well worth the time.
