I guess taking I40 all the way to Nashville and north on I24 is also an option if the time difference is reasonable.

You will be fine on either route. The one through Hurricane Mills/Cumberland City is the best road and by far the shortest. It is routinely used by semis. I live about halfway between Hurricane Mills and Clarksville, have driven the route hundreds of times.
Don't go all the way to Nashville. It's WAY too far, and you will miss some beautiful scenery.
