I used a loop of 1/2" vinyl tubing mounted on plywood for a DIY manometer. I also use the same tubing to level-in cabinets and counter tops. The meniscus of the water in the tube is extremely accurate (better than my construction based laser).
You'll want to seal off the heating ducts with cardboard and tape or throw a rug over them to eliminate air flow. Much of this is just common sense and if you miss a potential air flow spot, may not be critical. I know a lot of RV's have big cutouts for pipes and wires, some of those are inaccessible and air may escape. If the blower has enough air flow, sealing every air escape point isn't necessary. I would assume that a Sealtech operator doesn't spend much time trying to seal every escape point. The blower moves enough air to make up for the escape points.
