NFPA-58, the Liquefied Petroleum Gas Code adopted by most states as the basis for their own codes, does not appear to specify either way except in the case of those horizontal DOT cylinders that require filling in a vertical position, which usually requires dismounting them. The best answer for you would likely be to check with the LP dispensing station you intend to use and see what they require.
In Texas there is only one answer. They have to be removed to be refilled.

If you were on the East side of the metroplex I'd suggest Tractor Supply @ 1.89/gal (mesquite). They might remove the cylinders for you but I never expect the propane filler to remove empty or full cylinders for me. - Dutch_12078Explorer II

Well, you right about something - Easiest way is to remove them, take them to local propane dealer and have them filled.
