Can we assume you're stopping at the Casino in E St Louis? You sure don't want to be driving around any other part of ESL and definitely not after dark. Seriously. ESL is better than it use to be but the word 'better' is relative. There use to be 400+ murders a year in ESL. Now it's less than 200. Robberies, muggings, etc are still common. There are no businesses or tourist things in ESL. ESL is not a place you want to mess around. You'll find more trouble than you can imagine.
I-255 takes you right by E St Louis. If you're going to the Casino Queen then take I-255 to I-55 and then I-55 to 3rd St Exit. That will take you directly to the Casino.

This probably won't affect you directly, but Eastbound I64 is closed in STL this weekend. Here is a link explaining:

Also, there is Rams game this Sunday, and a large festival going on in Forest Park, called "Loufest". :R Both of these events are going to generate extra traffic.

Personally, I would still take 255 South and around, especially if you are towing.

In the 10+ times I've been through St Lou, half of the time I end up in the wrong lane that becomes an exit without any warning forcing me to take another scenic detour of downtown. Happened again this summer on the way out West as my GPS did not know about the new bypass coming in from the SE (I-255) and naturally I was in the wrong lane to take it and ended up going into town where I ended up in an exit lane and seeing downtown AGAIN. Unlike some I will not slow way down or force my way into a lane change at the last moment.
OP, I would take I-65 as it's newer, has longer sight distance, and has the exits marked better than I-70.
